The three main "sinners" causing caries are streptococcus mutans and sobrinus, and various species of lactobacilli. They were cultured and examined under the microscope. The microscopic methodes used are differential interference contrast (DIC) and gram-staining, along with some biochemical tests. The tests are not complete for detecting the exact species.
