bile acids are synthesized from cholesterol
KEY enzyme of bile acid synthesis is 7 Alpha hydroxylase which is a microsomal enzyme
primary bile acids are cholic acid and chenodeoxycholic acid
primary bile acids are conjugated with the help of glycine and taurine
Conjugated primary bile acids are deconjugated in intestine by intestinal bacteria.
Cholic acid is the deconjugated to deoxycholic acid while chenodeoxycholic acid is deconjugated to lithocholic acid
At alkaline pH of bile this bile acids exist as Sodium and potassium salts called as a bile salts.
bile salts play important role in in digestion of lipids
nuclear receptor that is bile acid binding receptor called as Farnesoid X receptor play a important role in suppresson of transcription of of gene of 7 Alpha hydroxylase.
when bile acid pool increase chenodexoxycholic acid bind with the farnesoid X receptor
and suppress 7 alpha hydroxylase and and so bile acid synthesis reduced.
FXR agonist like obeticholic acid is used in primary biliary cholangitis
