In development, hematopoietic stem cells (HSCs) pass through a temporal niche capable of regulating stem cell numbers through a variety of molecular and cellular mechanisms. Using multi-omic analyses, multifluorescence tagging and live imaging, we identify, at Harvard Medical School, a population of primitive macrophages that recognize newly formed hematopoietic stem and progenitor cells (HSPCs) and regulate stem cell clonality into adulthood. These macrophages detect surface-bound calr isoforms present on HSPCs and engulf the entire cell or remove small fragments of cellular material and promote expansion.
