The role of diabetes in increasing susceptibility, morbidity and mortality in cardiomyopathic heart failure is well established. In this talk, Dr Julieta Palomeque describes their work elucidating the changes in Ca handling in a prediabetic model in mice and rats. Their models show an increase in ROS production which activates CaMKII in the SR membrane which in turn phosphorylates RyR promoting further Ca release and arrhythmias. This SR Ca release is amplified as the mitochondria take up Ca from their shared microdomain, enhancing ROS production and CaMKII activation, and thus activating a vicious cycle.
