Noong ika-25 ng Mayo 1887, isinilang si Padre Pio sa isang simpleng pamilya ng masisipag na magsasaka sa Pietrelcina sa Timog Italya. Nag-aral siya sa ilalim ng pansariling guro hanggang sa pumasok sa nobisyado ng mga prayleng Capuchino sa gulang na 15. May mahinang katawan ngunit matatag namang kalooban, sa tulong ng biyaya ng Diyos ay nakatapos siya ng kailangang pag-aaral at inordinahang pari noong 1910. Manalangin tayo sa tulong ni Santo Padre Pio.
