The semen sample has to be obtained through masturbation after an abstinence of 3 to 5 days. The ejaculate is deposited in a collection pod. Once in the lab, the semen sample is prepered. Then, in order to select the sperm with the best motility, test tubes are used in which media of different densities are contained that allow for the separation of immotile semen, seminal plasma, cells of ditritus that are also present in the ejaculate. An initial centrifugation is performed and the sperm are collected with a pipette. Further rounds of washing and centrifugation are done after which the supernatant is discarded and the sediment is resuspended. The final sample obtained is evaluated under the microscope and finaly the sperm is stored in a test tube. The sperm that has been prepared is loaded into a syringe to which a catheter is attached. Then the speculum is introduced into the vagina allowing the entrance of the neck of the uterus to be made visible. The catheter that has been charged with sperm is introduced high into the uterus and the sperm ascends through the fallopian tubes arriving at the area where the oocyte is located and fertilisation of the oocyte is facilitated
