Preparation: Begin by preparing the Molisch reagent, which consists of a solution of alpha-naphthol in ethanol. To prepare the reagent, dissolve about 2 grams of alpha-naphthol in 100 mL of ethanol. This solution should be freshly prepared before performing the test.
Test setup: Take a small amount of the test sample (liquid or solid) in a test tube or a small flask. If the sample is solid, it should be finely ground and dissolved in water prior to the test.
Addition of reagent: Add a few drops (about 2-3 drops) of the Molisch reagent to the test sample. Mix the contents thoroughly by swirling the test tube.
Sulfuric acid addition: Carefully pour concentrated sulfuric acid along the sides of the test tube, allowing it to form a layer at the bottom. Tilt the test tube slightly to create a gradient between the acid layer and the sample solution.
Observation: Observe the appearance of a purple or violet ring at the interface of the two layers (sample and acid) within a few minutes. The purple ring indicates a positive result for the presence of carbohydrates.
It's important to note that the Molisch test is a qualitative test, meaning it only indicates the presence of carbohydrates but does not provide quantitative information. Additionally, the test may give false-positive results with certain compounds other than carbohydrates, such as phenols and aromatic amines. Therefore, it is often used as a preliminary test, and further confirmatory tests may be required for accurate carbohydrate identification
