The Phase II SWOG 1609 (DART) trial (NCT02834013) is investigating dual anti-CTLA-4 and anti-PD-1 blockade in dual anti-CTLA-4 and anti-PD-1 blockade in rare tumors. Young Kwang Chae, MD, MPH, MBA, National Cancer Institute, Bethesda, MD, shares the first results of ipilimumab and nivolumab in vulvar cancers. The primary endpoint was objective response rate (ORR) (RECIST v1.1) (confirmed complete (CR) and secondary endpoints included partial responses (PR); progression-free survival (PFS) and overall -survival (OS). Sixteen patients were evaluated, with an objective response rate (ORR) of 18.8% and a clinical benefit rate (CBR) of 31.3%. The median PFS was 2.2 months, and the median OS was 7.6 months. Adverse events included diarrhea, fatigue, pruritus, anorexia, and nausea. Correlative studies to identify response and resistance markers are ongoing. The study suggests potential benefits in vulvar cancers and calls for further exploration through expanded prospective studies.
