Various studies have shown that NAD+ levels decrease significantly during aging, and that restoration of NAD+ levels could help in extending lifespan and promoting health in animal models and in humans. Research indicates that supplementation with either NR or NMN can boost NAD+ levels. And in this video, I’m going to discuss the similarities and differences between these two NAD+ precursors, to figure out which one could be the best longevity supplement.
✅ NUMBER 1: EFFECTS OF NR OR NMN ON LONGEVITY?
NMN has been shown to increase lifespan in mice, which was one of the biggest discoveries when it comes to the science of aging. But surprisingly, recent data published in 2021 showed that NR failed to increase lifespan in mice. In this study, scientists fed the mice with NR at 1000 mg/kilogram of food using NR from Chromadex brand. Since they didn’t see any increase in lifespan, they finally assumed that NR might not be able to slow down aging, unfortunately.
✅ NUMBER 2: EFFECTS OF NR OR NMN ON INSULIN SENSITIVITY?
Let’s start with NR. NR has been shown to enhance glucose tolerance, improve insulin sensitivity and minimize weight gain in different studies using mouse models.
However, these results have not been replicated in humans, as NR supplementation at a high dosage of 2000 mg/day did not increase insulin sensitivity or glucose metabolism in obese patients.
Now, about NMN. NMN has also been shown to improve insulin sensitivity and lipid metabolism in mice. The good news is that the effect of NMN on insulin sensitivity was recently confirmed in humans, unlike NR. Scientists showed that taking 250 mg of NMN every day for 10 weeks improved insulin sensitivity in a group of women.
✅ NUMBER 3: EFFECTS OF NR OR NMN ON CARDIOVASCULAR HEALTH?
A study showed that NR supplementation attenuates the development of heart failure in mice.
However, scientists were not able to reproduce these promising results in humans with NR, as they didn’t see any improvement in cardiovascular health after NR supplementation. In this study, participants were taking NR at 1000 mg/day but didn’t notice any significant changes in blood pressure, body fat percentage, and exercise performance.
Now, regarding NMN, many groups showed the benefits of NMN on cardiovascular health in mouse models as well. But unlike NR, scientists were able to confirm the benefits of NMN on cardiovascular health in humans. First, a group investigated the effects of NMN on cardiovascular fitness in humans. They observed that taking NMN at 600mg/day could increase our aerobic capacity during exercise training. Another group showed that supplementing healthy old men with NMN at 250 mg/day only for 12 weeks could enhance NAD+ levels, muscle strength, and exercise performance as well.
✅ NUMBER 4:EFFECTS OF NR OR NMN ON BRAIN HEALTH?
Several studies showed that NR supplementation could improve cognitive function, decrease neuroinflammation and brain DNA damage in several mouse models of Alzheimer's disease.
Other studies demonstrated the neuroprotective role of NMN supplementation in mouse models as well. But no human studies have been done yet to confirm these data, neither for NR nor for NMN. So, we’re still waiting to see if these results can be translated to us.
✅ NUMBER 5: EFFECTS OF NR OR NMN ON FERTILITY?
A group demonstrated that NMN could counteract the negative effects of maternal age on fertility in a mouse model. These findings suggest that restoration of NAD+ levels could be an opportunity to rescue female reproductive function. Nothing has been done yet with NR in this field.
