These are the minimal steps I used for replacing a broken spoke in my rear wheel. I didn't have access to a truing stand this day, so I show how to do the repair without a truing stand. Depending on your bike, you may need to remove the cassette (or freewheel) and if you have disc brakes, possibly the rotor.
Depending on where the spoke is broken, you may have to remove the tire, tube and rim tape as well. On this particular wheel, I was able to replace the spoke and true the wheel without removing the tire. I was also able to complete the repair without a truing stand.
Spokes generally break either from something getting into the wheel or fatigue. The fatigue process is faster with a larger rider or carrying lots of weight.
