Video Summary:
Na+K+2Cl- cotransporter located at ascending limb of loop of Henle plays important role in not only Na+ but also water absorption. Its effect is multiplied by countercurrent multiplier. This in turn increases medullary osmolarity. Now this hyperosmolarity drives water absorption from distal tubule.
Mechanism of action of loop diuretics is inhibition of Na+K+2Cl- cotransporter. So all the effects are reduced. Final result is abolished medullary hyperosmolarity and reduced water reabsorption from DT. The unabsorbed water is lost in urine.
