The first in the list of "cosmetic analogs of Botox" is the peptide Argireline (INCI: Acetyl Hexapeptide-3). Appearing in the early 2000s, it quickly gained popularity. It is aimed at combating expression lines. The six amino acids of argireline repeat a region of the SNAP25 protein required for the binding of the synaptic vesicle of the axon to the presynaptic membrane. In the axon, argireline competes with the SNAP25 protein and instead integrates into the temporary SNARE protein complex. This complex is formed from several membrane proteins just before the vesicle binds to the membrane and is required for successful exocytosis. The defective complex cannot provide the necessary contact of the vesicle with the membrane, as a result, the release of the mediator into the synaptic cleft does not occur. This means that the muscle does not receive a signal to contract and continues to be in a relaxed state.
