Michael Deininger, MD, PhD, University of Utah, Salt Lake City, UT, discusses the role of membrane spanning 4-domains A3 (MS4A3), a trans-membrane protein, in tyrosine kinase inhibitor (TKI) resistance. Resistance to TKIs is associated with low MS4A3 expression in patients with chronic myeloid leukemia (CML), as MS4A3 promotes endocytosis of cytokine receptors. Downregulation of MS4A3 additionally shifts CML cells to an immature state, which enables resistance to TKIs.
